Evidenziando al contempo l\u2019inidoneit\u00e0, in tal senso, dei sistemi convenzionali.<\/p>\n<p>Le nuove tecniche di identificazione molecolare consentono infatti di eseguire uno\u00a0<em>screening<\/em>\u00a0pi\u00f9 accurato rispetto ai metodi colturali classici, caratterizzando al meglio proprio quei gruppi che possono contribuire alla diffusione dell\u2019antimicrobico-resistenza (AMR). Alcuni suoi ceppi, oltre a causare l\u2019insorgenza di malattie anche negli esseri umani, sono vettori di antimicrobico-resistenza. (2)<\/p>\n<p><strong>L\u2019antimicrobico-resistenza<\/strong>\u00a0\u00e8 sempre pi\u00f9 diffusa, in\u00a0<em>Salmonella<\/em>\u00a0come in\u00a0<em>Campylobacter<\/em>, con crescenti difficolt\u00e0 d\u2019impiego dei trattamenti farmacologici classici. (3) L\u2019utilizzo improprio degli antibiotici in medicina e veterinaria\u00a0ha portato a un incremento generale del fenomeno su scala globale, se pure le autorit\u00e0 europee (ECDC, EFSA, EMA, 2021) abbiano di recente osservata una lieve riduzione del\u00a0<em>trend<\/em>.<\/p>\n<h2><span class=\"ez-toc-section\" id=\"Sierotipi_di_Salmonella\"><\/span>Sierotipi di\u00a0<em>Salmonella<\/em><span class=\"ez-toc-section-end\"><\/span><\/h2>\n<p><strong>Un\u00a0sierotipo<\/strong>\u00a0(o\u00a0<em>serovar<\/em>) \u00e8 una sottospecie microbica che si caratterizza per un antigene specifico, a cui sar\u00e0 in grado di rispondere uno specifico anticorpo. La specie\u00a0<em>Salmonella enterica<\/em>\u00a0\u00e8 nota per i numerosi sierotipi di cui \u00e8 costituita e le diverse patologie che si possono sviluppare nell\u2019uomo. (4)<\/p>\n<p><strong>Il\u00a0sierotipo\u00a0<em>Reading<\/em><\/strong>\u00a0\u00e8 prevalente negli allevamenti di animali da reddito, in particolar modo tra i\u00a0bovini\u00a0e i tacchini. Sui bovini, in particolare, questo sierotipo sviluppa antimicrobico-resistenza con elevata frequenza &#8211; anche a causa di recenti epidemie diffuse in Nord America &#8211; ed \u00e8 ancor pi\u00f9 necessario trovare sistemi che ne facilitino l\u2019identificazione. (5)<\/p>\n<h2><span class=\"ez-toc-section\" id=\"Salmonella_la_tecnologia_CRISPR-SeroSeq\"><\/span>Salmonella, la tecnologia CRISPR-SeroSeq<span class=\"ez-toc-section-end\"><\/span><\/h2>\n<p><strong>La sorveglianza<\/strong> e l\u2019isolamento\u00a0di\u00a0<em>Salmonella<\/em>\u00a0generalmente\u00a0dipendono dal metodo colturale impiegato. Questo approccio attualmente permette di individuare pochi sierotipi &#8211; soprattutto, quelli pi\u00f9 abbondanti all\u2019interno della popolazione analizzata &#8211; e spesso non considera quelli minoritari, i quali tuttavia operano come serbatoi di AMR. (6)<\/p>\n<p><strong>CRISPR<\/strong>\u00a0(<em>Clustered Regularly Interspaced Short Palindromic Repeats<\/em>, utilizzata anche per l\u2019<em>editing<\/em>\u00a0genomico,\u00a0come si \u00e8 visto) &#8211; nello specifico, CRISPR-SeroSeq &#8211; \u00e8 una tecnologia che invece permette di identificare le frequenze relative a pi\u00f9 sierotipi in un solo campione. Ed \u00e8 stata perci\u00f2 sperimentata anche per identificare i sierotipi di\u00a0<em>Salmonella<\/em>. (7)<\/p>\n<h2><span class=\"ez-toc-section\" id=\"CRISPR-SeroSeq_risultati_degli_studi\"><\/span>CRISPR-SeroSeq, risultati degli studi<span class=\"ez-toc-section-end\"><\/span><\/h2>\n<p><strong>L\u2019analisi\u00a0<\/strong>delle popolazioni isolate da matrici di origine bovina ha permesso di identificare che due terzi dei campioni positivi alla\u00a0<em>Salmonella<\/em>\u00a0esaminati contenevano numerosi sierotipi tra i quali il\u00a0<em>Reading<\/em>\u00a0(anche se presente in minoranza, probabilmente perch\u00e9 poco competitivo).<\/p>\n<p><strong>La\u00a0tecnologia CRISPR-SeroSeq\u00a0<\/strong>\u00e8 effettivamente risultata in grado di classificare in modo pi\u00f9 preciso e accurato le popolazioni di\u00a0<em>Salmonella<\/em>\u00a0presenti e di individuare numerosi sierotipi, anche quelli difficilmente individuabili come il sierotipo <em>Reading<\/em>, soprattutto con i metodi di isolamento tradizionali e i relativi ceppi AMR.<\/p>\n<p><strong>Il\u00a0trattamento con antibiotici<\/strong>\u00a0ha mostrato una variazione di sviluppo e comportamento delle popolazioni isolate, di particolare utilit\u00e0 per identificare i sierotipi e ceppi pi\u00f9 sensibili e quelli pi\u00f9 resistenti ai vari trattamenti.<\/p>\n<h2><span class=\"ez-toc-section\" id=\"Conclusioni_provvisorie\"><\/span>Conclusioni provvisorie<span class=\"ez-toc-section-end\"><\/span><\/h2>\n<p><strong>La funzionalit\u00e0<\/strong>\u00a0della nuova tecnologia per la classificazione delle specie di\u00a0<em>Salmonella<\/em>\u00a0in matrici animali \u00e8 sicuramente un nuovo strumento utile per effettuare uno\u00a0<em>screening<\/em>\u00a0pi\u00f9 efficace e accurato non solo del microrganismo, ma di tutti i portatori di antimicrobico-resistenza.<\/p>\n<p><strong>Il sistema<\/strong>\u00a0dovr\u00e0 venire testato anche in altri contesti, in vista della sua validazione.
